About the PhD:

The current PhD study investigated the protective role of extracellular vesicles and miRNAs from remote ischemic conditioning in cellular model of stroke. The project was carried out by Katrine Tang Stenz, who is defending her dissertation on 07/07 2023.

Stroke is one of the leading causes of deaths and disabilities world wide. Acute ischemic stroke (AIS) is characterized by a sudden occlusion of a blood vessel, is the most common type of stroke and treatment is based on removal of the blood clot. Remote ischemic conditioning (RIC) has been suggested as an additional treatment to traditional stroke treatments, as it may confer protection of the neurons and vasculature of the brain. The aim of this PhD project was to investigate the underlying molecular mechanisms of RIC. This was done in primary human brain microvascular endothelial cells (HBMECs) – cells which are a critical component of the blood brain barrier. Nano-sized extracellular vesicles (EVs) were isolated from plasma samples from healthy volunteers which underwent RIC. Furthermore, miRNAs were identified in EVs. Both EVs and miRNAs were tested for the protective effect in a cellular model of AIS. After testing the protective effect of RIC derived miRNAs, the gene expression of HBMECs were investigated using RNA sequencing.
